Why is Competitive Play locked for me if I played Overwatch 1?

Players who reached Level 25 in the original game receive automatic access via account merging. All other players must fulfill the 50-win challenge requirement to unlock Competitive.

Do Arcade or Custom Game wins count toward the 50 wins?

No, those don’t count. Only wins in Unranked Quick Play (Role Queue or Open Queue) contribute to the Competitive Play Qualification challenge.
